The raw material is defatted and dehydrated with acetone or ethanol, soaked with distilled water, filtered, and then treated with sodium chloride aqueous solution and chloroform solution. Trypsin is then added and kept warm to obtain a mixed solution, which is finally treated with an ion exchanger and purified to obtain refined hyaluronic acid. The extraction rate of this method is extremely low, only about 1%, and the separation process is complicated, which makes hyaluronic acid expensive, up to US$5,000 per kilogram, limiting its large-scale use in cosmetics.

2. Microbial fermentation : using glucose as the carbon source in the fermentation liquid. Ferment in the culture medium for 48 hours. After the fermentation is completed, the mycelium and impurities are filtered out, and then a high-purity product is obtained by simple operations such as alcohol precipitation. The advantage of hyaluronic acid produced by the fermentation method is that the molecular weight can be set according to the product design. The key to the fermentation method lies in the selection of bacteria, with streptococci, lactococci, etc. being the most commonly used.
