Urine protein positive 2+ is the main indicator in our urine routine examination. It is very important for us to analyze diseases such as urethritis and vaginitis. If the urine protein test results are all 2+ throughout the day, it may be because our kidneys are diseased. At this time, we should pay attention to it and cooperate with the doctor's treatment in time. Let us now learn how to judge the results of urine protein examination.

Positive urine protein means that the results of a routine urine test or a 24-hour urine protein quantitative test show “+”.

There is always a certain amount of protein in the blood that is indispensable to human life activities. Some of the protein is filtered into the urine in the glomeruli of the kidneys, but is absorbed back into the blood in the renal tubules. Therefore, if the kidneys function normally, only a small amount of protein will appear in the urine. However, when there is an obstruction in the kidneys and ureters, a large amount of protein will leak out and become proteinuria. There is a trace amount of protein in the urine of normal people, which is qualitatively negative within the normal range and recorded as (-). When the protein content in urine is more than 0.15g/24h, it is called proteinuria, and routine urine tests may show positive results. The normal 24-hour urine protein range is ≤0.15g, and routine laboratory tests are negative. If the urine protein test is > 150 mg/day, that is, the urine protein is positive, it means that the amount of urine protein excreted by the human body has increased significantly, which is abnormal urine protein.

Persistent positive urine protein often indicates kidney disease. Therefore, clinically, the degree of kidney damage and the effectiveness of kidney disease treatment can be determined based on the amount of positive urine protein. Therefore, if abnormal urine protein occurs, it must be effectively controlled and eliminated to prevent the disease from worsening and progressing.

The method of indicating the strength of urine protein positivity is generally to detect the protein content in urine through a 24-hour urine protein quantitative test. There are three test methods: test paper method, sulfosalicylic acid method and heated acetic acid method. Both the sulfosalicylic acid method and the heated acetic acid method are based on the turbidity reaction and define the absence of turbidity or precipitation as negative (-), and the presence of turbidity or precipitation as positive (+). The sulfosalicylic acid method is easy to operate and has high sensitivity, and can be widely used in censuses. However, its sensitivity to albumin is higher than that to globulin, and there are many influencing factors, which can easily cause false negatives or false positives. The heated acetic acid method has basically the same sensitivity to albumin and globulin, with fewer influencing factors and higher accuracy. The urine protein content was estimated based on the turbidity reaction as follows:

Urine protein <0.1 g/L: -;

Urine protein is 0.1-0.2 g/L: ±;

Urine protein is 0.2-1.0 g/L: +;

Urine protein is 1.0-2.0 g/L: ++;

Urine protein is 2.0-4.0 g/L: +++;

Urine protein>4.0g/L: ++++.

Normal urine contains trace amounts of protein (24-hour urine protein quantity <150 mg), which cannot be detected by ordinary detection methods and the test result is negative. If it exceeds this range, it can be detected and is called proteinuria. However, since the urine protein test only measures the result of one urine test, it is easily affected by the concentration and dilution of the urine and often cannot accurately reflect the degree of proteinuria. When the human body excretes strenuous exercise, heavy physical labor, emotional excitement, excessive cold, overheating, or is under stress, the amount of urinary protein excretion may increase. This is called transient proteinuria and will return to normal after a few hours or days.
