The occurrence and development process of the lesion is followed, and finally a pathological diagnosis is made.

Tissue sampling is obtained from the human body (surgical resection specimens, biopsy specimens, autopsy specimens) or animals according to the specific requirements of teaching, scientific research and external examination, and the location and method of sampling are determined. The HE staining procedure is as follows: dewaxing, debenzening, rehydration, staining, dehydration, transparency, and sealing. The contrast stain in conventional hematoxylin staining is eosin. In recent years, some laboratories in the UK and the US have used phloxine. In addition, Orange G, Biebrich scarlet, Bordeaux red, etc. are also used as contrast stains. Eosin is a commonly used dye for staining cytoplasm, collagen fibers, muscle fibers, eosinophilic granules, etc.
