The pathogenesis of pituitary tumors is a complex multi-step process involving multiple factors, which has not yet been clarified. There are mainly two hypotheses: one is the mechanism of abnormal hypothalamic regulation, and the other is the mechanism of pituitary cell defects. The former believes that the cause originates from the hypothalamus. Under the abnormal regulation of the hypothalamus, it causes hyperpituitarism, hyperplasia and adenoma; pituitary adenoma is just one of the manifestations of hypothalamic-pituitary dysfunction. The latter believes that local factors in the pituitary gland cause the pituitary cells to be hyperfunctional, thus forming adenoma. In recent years, with the continuous advancement of molecular biology technology, research on the correlation between gene mutations and the occurrence of pituitary tumors has continued to deepen. At present, it is relatively clear that the gsp oncogene is closely related to pituitary GH adenoma. The gsp gene is a new proto-oncogene formally defined by the mutation of Gsαa. The Gsαa gene is an independent gene sequence of about 20Kb, consisting of 13 exons and 12 introns. Its base sequence and structural function have been basically clarified, and its point mutation leads to the mutation of the Gs protein αa2 subunit (Gsαa). Gs protein is a member of the G protein family. Its function is to transmit stimulation signals from cell surface receptors to the catalytic unit of adenylate cyclase to promote the synthesis of cyclic adenosine monophosphate (cAMP). Since the secretion of growth hormone is cAMP-dependent, Gs protein is closely related to the secretion of GH; and more and more evidence shows that cAMP is also a growth medium. In this way, changes in Gs protein may be directly or indirectly related to tumor growth. Many clinical studies have found that gsp mutations are more common in patients with pituitary GH adenoma, and this mutation is only found in tumor cells but not in surrounding cells, and the mutation has obvious activity. These results indicate that there may be specific gsp oncogene mutations in the occurrence and development of pituitary GH adenoma. We know that although pituitary tumor is a chronic disease, it will gradually develop. When the disease develops to a certain extent, it will cause great harm to the patient's physical and mental health. Therefore, patients must not be careless after suffering from this disease. They should receive timely treatment to minimize the harm caused by this disease. |
