The rabies virus is a RNA-type rhabdovirus, with one end convex and the other concave, shaped like a bullet, with a diameter of 65-80nm and a length of about 130 to 240nm. The virus can be easily inactivated by sunlight, ultraviolet light, formaldehyde, mercuric chloride quaternary ammonium compounds (such as Sanisol), lipid solvents, 50%-70% alcohol, etc. The suspension can be inactivated by heating at 56°C for 30-60 minutes or at 100°C for 2 minutes. The virus can remain active for several years at -70℃ or at 0-4℃ after freeze-drying. Infected tissue can be stored in 50% glycerol for testing. Rabies virus contains five proteins, namely glycoprotein (G), nucleoprotein (N), dimerase (L), phosphoprotein (NS) and matrix (M). The latter two are small molecule proteins. G can lead to the formation of neutralizing antibodies in the body, which can fight against viral attacks. N produces antibodies that are not neutralizing and can be used to detect intraplasmic inclusion bodies. Rabies virus has two main antigens: one is the glycoprotein antigen on the outer membrane of the virus, which can bind to the acetylcholine receptor to make the virus neurotoxic and produce neutralizing antibodies and hemagglutination inhibitory antibodies in the body. The neutralizing antibody has a protective effect; the other is the nucleoprotein antigen on the inner layer, which can produce complement binding antibodies and precipitins in the body, but has no protective effect. Viruses isolated from patients and diseased animals are called natural viruses or street viruses. They are characterized by strong virulence, but after passing through the rabbit brain many times, they become fixed viruses with reduced virulence and can be used to make vaccines. The rabies virus is not heat-resistant and can be inactivated in 15 to 30 minutes at 56°C or 2 minutes at 100°C. It is sensitive to disinfectants such as acids, alkalis, chlorhexidine, and formalin. Sunlight, ultraviolet rays, ultrasound, 70% alcohol, 0.01% iodine solution, and 1%-2% soapy water can also inactivate the virus. |
