How to store CST antibodies?

A: Please store the original tube of antibody at -20°C and do not aliquot. Because the antibodies are stored in a buffer containing 50% glycerol, they remain liquid at -20°C and will not degrade due to repeated freezing and thawing. Please refer to the product instructions for specific storage temperature.

2. Question: How to store SDS-lysed phosphorylated protein samples?

A: If used in the short term (less than 3 months), SDS sample cell lysate can be stored at -20°C. If stored long term, it should be stored at -80°C. The degradation of activated protein samples varies greatly, with the rate of degradation depending on the nature of each protein and how much activated protein is present in the sample.

3. Question: Can I incubate the antibody at room temperature for 1 hour instead of overnight at 4°C?

Answer: Not recommended. CST's activation state-specific antibodies are designed to distinguish between one or two post-translationally modified amino acid residues. These modifications include phosphorylation, acetylation and enzymatic cleavage. Most antibodies directed against these modification sites have lower affinities compared to antibodies recognizing the total protein. Therefore, it is critical to prolong the reaction time of antigen and antibody by incubating at 4°C overnight to obtain the best signal-to-noise ratio.
